| Step 1. Homogenization | Before each use, the turbidity standard tube should be vigorously shaken on a rotary mixer. Visually inspect the tube to ensure the turbidity is uniform. |
| Step 2. Preparation | Add physiological saline to the blank tube and sterilize it for later use. |
| Step 3. Suspension | Using aseptic technique, transfer a single colony of the pure culture to be tested into the prepared physiological saline tube and suspend it. |
| Step 4. Comparison | Using a color chart as a background, visually compare the turbidity until the concentration matches that of the required standard tube. |
Precautions:
1. If large particles appear, this standard tube should be replaced.
2. This turbidity tube is only for rough estimation of bacterial count.
3. Before each use, the turbidity standard tube should be vigorously shaken on a vortex mixer, and its appearance should be visually inspected to ensure uniform turbidity.
